Tirzepatide Peptide

Tirzepatide is a dual agonist of the glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) receptors. Tirzepatide is a linear polypeptide made up of 39 amino acids, Tirzepatide has been chemically modified by lipidation to potentially improve its stability.

Tirzepatide Research

Research indicates that Tirzepatide may act identically to GIP at GIP-R, but its activity at GLP-1R appears to favor cAMP production over β-arrestin recruitment. Tirzepatide has also been suggested by researchers to modulate adiponectin levels, and may potentially act on lipoproteins to reduce triglycerides, apoC-III, and others. The peptide may potentially increase in the expression of eNOS, an enzyme generating nitric oxide, and may reduce NF-κB signaling, decrease MMP-9 activity, inhibit inflammatory cytokine synthesis, and reduce macrophage activity.

A study explored the potential of GIP overexpression in heterozygous transgenic (Het GIP Tg) murine models in comparison to Wild Type (WT) mice. Upon peptide exposure, the observations made on the Het GIP Tg mice suggesting enhanced β-cell function, as well as markedly less macrophage infiltration and hepatic steatosis than seen in WT mice. This could potentially be linked to coding for ATP citrate lyase (Acly) and interleukin 4 receptor alpha (IL4ra).
